METHOD FOR PROMOTING SEEDLING STRENGTHENING BY PHYSICAL STIMULATION
OF CUCUMBER COTYLEDON Technical Field The present invention belongs to the technical field of cucumber seedling growing, and particularly relates to a method for promoting seedling strengthening by physical cotyledon stimulation of cucumber.
Background Cucumber is an important vegetable crop. The scale and yield of cucumber in China rank first in the world. The cucumber plays an important role in structural adjustment of agriculture and increase of farmer income. Plug seedling growing of cucumber may achieve advantages of increasing seedling growing efficiency, saving seeds and space, bringing convenience to management, facilitating pest control and performing large-scale production, and is widely applied to modern facility agriculture.
For the traditional plug seedling growing technology, seedlings are mainly strengthened by spraying chlormequat chloride, mepiquat chloride, B9 and the like in production, or hormones are directly applied to nursery substrates, thereby causing hormone poisoning of seedlings, and harm of residual poisons to soil, environment and next-stubble crops. To overcome the above defects, a green and safe seedling strengthening method is in an urgent need.
Summary To overcome defects of the prior art, the present invention provides a method for promoting seedling strengthening by physical cotyledon stimulation of cucumber. According to the present invention, cucumber seedlings are strengthened in a manner of performing physical cotyledon stimulation on the cucumber, and cultivated cucumber seedlings are better than conventional seedlings growing in the aspects of biomass, root activity and seedling strength index.
Technical solutions of the present invention are as follows: The method for promoting seedling strengthening by physical cotyledon stimulation of cucumber includes the following steps: performing physical cotyledon stimulation after induced germination when the cotyledons coming out of the shoot tip developed, i.e. after the cotyledons coming out of the shoot tip flattened, wherein the physical stimulation includes an operation of performing physical disturbance at an optional position of unilateral cotyledon or removing partial cotyledon within 1/2 of leaf vein length of a single unilateral cotyledon perpendicular to the main leaf vein of the cotyledon; placing cucumber seedlings on a cool lee side until the wound heals after physical stimulation; and continuing to cultivate the cucumber seedlings after the wound heals.
As a preferred solution, when the cotyledon coming out of the shoot tip developed, partial cotyledon within 1/8-3/8 of leaf vein length of a single unilateral cotyledon is removed perpendicular to the main leaf vein of the cotyledon.
Further, when the cotyledon coming out of the shoot tip developed, partial cotyledon within 1/4 of leaf vein length of a single unilateral cotyledon is removed perpendicular to the main leaf vein of the cotyledon.
As a preferred solution, after physical stimulation, the cucumber seedlings are placed on the cool lee side for 1-2 hours.
As a preferred solution, the seedling growing environment is as follows: a light period is 12h/12h, a diurnal temperature is 28°C/18°C, daytime light intensity is 600 umol-m2-s! (where 18 pumol-m2-s' = 1000 Ix), and air humidity is 55-65%; and the seedlings are watered with clear water and nutrient solution before two leaves and one bud develop, and the seedlings with cotyledons are watered with total nutrient solution after two leaves and one core are developed until seedling growing is ended.
As a preferred solution, after induced germination, the cucumber is seeded in an empty plug filled with nursery substrate.
Further, the nursery substrate is a substrate prepared by mixing turf, vermiculite and perlite according to a volume ratio of 3:1:1.
The present invention has beneficial effects as follows:
1. According to the present invention, by removing the partial cotyledon in the flattening period of the cucumber cotyledon to perform physical stimulation, endogenous hormone production is facilitated, and root and seedling strengthening is promoted; and problems such as toxication and pollution caused by exogenous hormone application in the cucumber seedling growing process are solved; the method is green and environment-friendly, and cost is saved.
2. The cucumber seedlings cultivated by the method in the present invention are better than conventional seedling growing in the aspects of the biomass, root activity and seedling strength index.
3. The method in the present invention is simple in operation, easy to master and suitable for plug seedling growing of vegetables.
Description of Drawings To clearly illustrate technical solutions in embodiments of the present invention or in the prior art, drawings to be used in the description of embodiments or the prior art are simply introduced below. Apparently, the drawings in the description below are some embodiments of the present invention only. Other drawings may be obtained by those ordinary skilled in the art according to the drawings on premise of not making creative work.
Fig. 1 is a diagram of influence of different degrees of physical cotyledon stimulation degrees on plants.
Detailed Description A method for promoting seedling strengthening by physical cotyledon stimulation of cucumber includes the following steps: performing physical cotyledon stimulation after induced germination when the cotyledons coming out of shoot tip developed wherein the physical stimulation includes an operation of performing physical disturbance at an optional position of unilateral cotyledon or removing partial cotyledon within 1/2 of leaf vein length of a single unilateral cotyledon perpendicular to the main leaf vein of the cotyledon; placing cucumber seedlings on a coal lee side until the wound heals after physical stimulation; and continuing to cultivate the cucumber seedlings after the wound heals.
The physical disturbance is performed at an optional position of the unilateral cotyledon, wherein the physical disturbance includes an operation of filing a hole or cutting one or more seams on the unilateral cotyledon; the area of the hole is smaller than 1/2 of the area of the whole cotyledon; the operation of cutting one or more seams is that the cotyledon does not drop from the cutting position before the main leaf vein is cut.
As a preferred solution, the seedling growing environment is as follows: a light period is 12h/12h, a diurnal temperature is 28°C/18°C, daytime light intensity is 600 umol-m™-s™*, and air humidity is 55-65%; and the seedling is watered with clear water and nutrient solution before to two leaves and one bud develop, and the seeding with cotyledons are watered with total nutrient solution after two leaves and one bud developed until seedling growing is ended.
As a preferred solution, after induced germination, the cucumber is seeded in an empty plug filled with nursery substrate.
Further, the nursery substrate is a substrate prepared by mixing turf, vermiculite and perlite according to a volume ratio of 3:1:1 Application examples: 1) Accelerating germination and seeding growth to cotyledon flattening including steps: selecting full seeds, soaking the seeds in warm water at about 50°C, taking out the seeds within 6 h, sucking surface moisture, wrapping the seeds by wet gauze, performing germination induction at 28°C, and seeding the seeds in a 50-well plug filled with a nursery substrate (a ratio of turf to vermiculite to perlite is 3:1:1) after the seeds become white.
A cultivation environment is as follows: a light period is 12h/12h, a diurnal temperature is 28°C/18°C, daytime light intensity is 600 umol-m-s™, and air humidity is 60%. After 7 days from accelerating germination seeding of cucumber, physical stimulation may be performed when the cotyledon coming out of the shoot tip flattens. 2) Physical stimulation of different degrees Fig. 1 respectively shows plant growth situations in the same cultivation environment from the left to right for a cucumber seedling of normal growth without any treatment, a cucumber seedling subjected to mechanical stimulation of a unilateral cotyledon in the cotyledon flattening period (a seam of not longer than one half of the cotyledon width is cut on the unilateral cotyledon), a cucumber seedling of which 1/4 of leaf vein length of a single unilateral cotyledon is removed perpendicular to the main leaf vein of the cotyledon in the cotyledon flattening period, a cucumber seedling of which 1/2 of the leaf vein length of the single unilateral cotyledon is removed perpendicular to the main leaf vein of the cotyledon in the cotyledon flattening period, a cucumber seedling of which 3/4 of the leaf vein length of the single unilateral cotyledon is removed perpendicular to the main leaf vein of the cotyledon in the cotyledon flattening period, and a cucumber seedling of which the single unilateral cotyledon is totally removed in the cotyledon flattening period.
Treatment to cucumber seedlings after physical stimulation: including steps: placing the cucumber seedlings on a cool lee side after physical stimulation; and continuing to cultivate the cucumber seedlings in a normal seedling growing environment after the wound heals (about 1 hour), wherein the light period is 12h/12h, the diurnal temperature is 28°C/18°C, the daytime light intensity is 600 ymol-m2-s!, and the air humidity is 60%. The cotyledon is watered with clear water before flattening, watered with 1/2 nutrient solution while flattening to two leaves and one core and watered with total nutrient solution after flattening to two leaves and one core.
The seedling can be transplanted for field planting while growing to four leaves and one core.
It may be seen from Fig. 1, compared with CK control, by virtue of mechanical stimulation of T1 unilateral cotyledon (a seam not longer than one half of the cotyledon width is cut on the unilateral cotyledon) and removal of 1/4 of T2 unilateral cotyledon, growth of plants can be improved, the leaf area of the plants is increased, and growth is stronger.
Moreover, the effect is the best when 1/4 of the unilateral cotyledon is removed.
Compared with CK, removal of 1/2 of T3 unilateral cotyledon does not have great change.
In addition, overstimulation of T4 and T5 (removal of more than 1/2 of the leaf vein length of the single unilateral cotyledon) may cause plant growth inhibition, growth vigor of the cucumber seedlings is poorer than that of conventional breeding, and removal of more than 1/2 of the leaf vein length of the single unilateral cotyledon is not recommended.
